Understanding the establishment of cellular identity is a major goal of modern biology. Programming and reprogramming cellular identity elicits tremendous scientific and public interest. The groundbreaking work of Takahashi and Yamanaka in 2006 established a landmark in cellular reprogramming with the generation of induced pluripotent stem cells (iPSCs) by forced expression of just four transcription factors. Similar to embryonic stem cells, iPSCs can proliferate and self-renew indefinitely under appropriate conditions and give rise to all cell types in the body, which gives these cells great potential for use in regenerative medicine. I will present a new strategy that can easily generate iPSCs from adult somatic cells using only a chemical cocktail, a procedure that is termed “chemical reprogramming”. Our study provides an important proof-of-principle demonstration that manipulation of signaling pathways and chromatin structure alone are sufficient to endow cells with pluripotency. This finding may lead to a safe and simple way to generate patient-specific iPSCs without the need for viral infection or introduction of other transgenes.
