摘要:

Cancers require two main elements to initiate: the genetic events that drive tumorigenesis, and a cell permissive to tranformation by the genetic driver, the cell of origin.  Using pancreatic ductal adenocarcinoma (PDA) as a model, I show that a major determinant of permissivity to transformation is the existence of a core transcriptional network amenable to transformation in response to specific oncogenic signals. In support of this, I show that In PDA, mutant Kras signaling co-opts a pancreatic stemness transcriptional network to drive its oncogenic gene expression program. I go on to show that lineage-specific tumor suppression by TGF-β involves disruption of this gastrointestinal-specific transcriptional network through the induction of a lethal epithelial-to-mesenchymal transition.
