摘要:

As a decisive step in the secretory pathway, protein translocation has been studied extensively for over 30 years. However, some of the fundamental questions remained unsolved, e.g. how highly diverse signal sequences of secretory proteins are recognized by the protein translocation channel and how protein translocation is driven by ATP hydrolysis. To answer these questions, I developed an in vivo system to assemble a translocation machinery that consisted of all three components required for protein translocation, the SecY channel, the SecA ATPase motor, and a peptide substrate that is being translocated. I determined the crystal structure of the machinery. The study uncovered a universal recognition mechanism of signal sequences.
