Proper selection of the translation initiation site on mRNAs is crucial for the production of desired protein products. In eukaryotes, ribosomal scanning is a well-accepted model for start codon selection.  It is commonly assumed that the first AUG codon that the scanning ribosome encounters serves as the start site for translation. However, one or more potential initiation sites could exist upstream or downstream of the main start codon. We developed quantitative profiling of initiating ribosomes, which revealed a surprising variety of alternative start codons.  The molecular mechanism responsible for the choice of alternative start codons is complex. In addition to the sequence context surrounding the start codon, we found that mRNA methylation in the form of m6A influences start codon selection. Dysregulation of this pathway has been implicated in human diseases such as diabetes and cancer. How mRNA methylation controls alternative translation represents an exciting research field to be fully exploited.
