Synaptic transmission is maintained by a delicate sub-synaptic architecture and even mild alterations in synapse structure drive functional changes during experience-dependent plasticity and pathological disorders. Key to this architecture is how the distribution of vesicle fusion sites at presynaptic terminals corresponds to the position of receptors on the postsynaptic neurons. Despite long recognition of its significance, this has not been precisely described, due in part to the limited resolution of light microscopy.  By adapting the super-resolution localization microscopy and developing extensive new analysis approaches, Dr. Tang performed a quantitative analysis of nanoscale “anatomy” on synapses between cultured hippocampal neurons and discovered a trans-synaptic molecular “nanocolumn” in which active zone regions with the highest likelihood of release are aligned to the densest receptor areas. More importantly, these aligned nanoscale subdomains are actively reorganized during synaptic plasticity. This architecture suggests a simple organizational principle of central nervous system synapses to maintain and modulate synaptic efficiency.
