Abstract:
Post-transcriptional RNA regulations play critical roles during development and in cancers, yet knowledge about which remains limited in embryonic stem cells (ESCs). My recent study discovered two cis-acting sequences within a pri-miRNA can selectively and dynamically regulate expression of mature miRNAs in ESCs. Cleavage of pri-miRNA produces a miRNA biogenesis intermediate that we termed ‘progenitor-miRNA’ (pro-miRNA). Pro-miRNA biogenesis specifically requires a complex containing CPSF3 and ISY1 proteins. By further investigating the biological relevance of pro-miRNA pathway, I find that dynamic expression of ISY1 gene during the naïve to primed ESC transition defines a unique phase of ‘poised’ pluripotency characterized by distinct miRNA and mRNA transcriptomes and the ability to contribute to mouse chimeras. ISY1 promote exit from the naïve state and is necessary and sufficient to induce and maintain poised pluripotency, which is an indispensable intermediate for the naïve to primed cell transition. This discovery provides an explanation for the early embryonic lethality of global miRNA deficiency known for a decade.
